Effect of WIN55, 212-2 on lowing rabbit IOP

OBJECTIVE: Glaucoma is one of causes of blindness clinically with the damage of optic papillary and retinal never fiber layers and glaucomatous visual field loss. To reduce the intraocular pressure( IOP) is a main purpose of management of glaucoma. The aim of this paper was to assess the effect of WIN55, 212-2 on IOP in rabbit with carbomer-induced chronic elevation of IOP. METHODS: Experimental ocular hypertension model was induced in 24 right eyes of 24 rabbits by the anterior chamber injection of complex carbomer. 24 left eyes were in blank control group. 0. 9% NS,0. 5% timolol and 0. 125% WIN55, 212-2 ophthalmic drops was used topically 1 week after injection respectively in 3 number-matched groups. IOP was detected once per day after administration of the ophthalmic formulations for 3 weeks. RESULTS: The IOP was(34. 3 ±2. 6) mmHg in timolol group, (34. 6 ±3. 1 ) mmHg in WIN55, 212-2 group and (34. 1 (plus or minus) 2. 5 ) mmHg in NS group, showing a significant increase in comparison with blank control group(15.4 ±3. 9) mmHg (P < 0. 05 ). IOP reduced rapidly to 65% in WIN55, 212-2 group and 78% in timolol group 1 weeks after utilize of drugs, and decreased to 45% and 60% respectively in WIN55, 212-2 group and timolol group 3 weeks after administration of drugs. The maximum reduction of IOP was significantly different among the groups (P <0.01). CONCLUSION: These results indicate that WIN55, 212-2 solution has a in longer action duration and good effectiveness in lowering IOP. LA: Chinese
