Effects of two forskolin analogues on the elevation of ocular pressure in human

OBJECTIVE: To investigate the effects of two compounds, isoforskolin (isoF) and deacetylforskolin (deaF) on the acute increase of intraocular pressure (IOP) induced by water-drinking (14 ml/kg) in healthy subjects when topically administered. METHODS: The study was carried out in double-blind and randomized healthy subjects. Drops of the two drugs were instilled into one eye while the corresponding vehicle was administered to the contralateral eye. IOP was measured with pneumatonometer before and after administration respectively. RESULTS: The results showed that both isoF (0.5%) and deaF (1.0%) decreased the acute elevation of IOP induced by water-drinking with the maximal inhibitory rates of 11.4 and 12.0%, respectively. The drugs were well tolerated by subjects. CONCLUSION: IsoF and deaF are effective for the inhibition of IOP increase.
