Immunofluorescence-histochemical changes of the astrocytes in cribriform plate due to high intraocular pressure in rats

AIM: To observe the expression of glial fibriliary acidic protein (GFAP) and Nogo-C in astrocytes of optic nerve head in cribriform plate region in rats, including glaucoma or normal eyes. METHODS: The experiment was conducted at the Department of Ophthalmology and the Third Research Room, Institute of Battle Surgery, Daping Hospital, Third Military Medical University from March to July 2004. PROCEDURE: 1. The right eyes of 16 normal SD rats performed light coagulation at the trabecular meshwork striding over limbus corneae with 532-diode laser. 2. The changes of intraocular pressure (IOP) was measured with Tonopen tonomet. 3. The immunofluorescence-histochemistry and laser-confocal microscopy were utilized to observe the expression of GFAP and Nogo-C of astrocytes. RESULTS: Sixteen eyes in the model group and 16 eyes in the non-model group were all involved in the result analysis. 1. Condition of IOP in the model group and the non-model group: After laser light coagulation, the IOP in the model group showed a trend of increase, and it was significantly higher on the third day than that in the non-model group (3.12 ± 0.01, 2.29 ± 0.03 kPa, P < 0.01). It lasted 60 days and all obviously higher than that in the non-model group (3.29 ± 0.03, 2.30 ± 0.02 kPa, P < 0.01). 2. Results of immunofluorescence-histochemical staining on eyeball sample in the model group and non-model group: Sixty days after high IOP, the expression of GFAP and Nogo-C of astrocytes in cribriform plate region of optic papilla was powerful with high density. The red fluorescence of GFAP, green fluorescence of Nogo-C and the yellow fluorescence (gray value) after the stack of red and green in the astrocytes in the model group were significantly higher than those in the non-model group (19.13 ± 0.12,17.82 ± 0.23, 18.30 ± 0.14 vs 3.11 ± 0.15, 6.52 ± 0.16, 4.16 ± 0.20, P < 0.01). CONCLUSION: Astrocytes may take part in the furt her lesion of nerve fiber in glaucoma, and inhibit the regeneration of nerve fiber axis-cylinder. LA: Chinese
