OBJECTIVE: Research has proved thai the apoptosis of retinal ganglion cells is closely link to the retinal excitability toxicity induced by glutumate, and glutamate plays an important role in ihe pathogenesis and progress of glaucoma. Present study was to explore the effect of hypericin , a specific protein kinase C antagonist, on relinal glutamate content in chronic ocular hvperiension rabbits. METHODS: Thirty-two rabbits were used to establish chronic ocular hypertension model by posterior chamber injection of alpha-chyniotrypsin to elevate the intraocular pressure (IOP) to 30 mmHg or more. Ten days after the injection. 0. 1 ml hypericin or saline was injected into the vitreous cavity in established 24 rabbits model. Three rabbits from the each group were sacrificed for relinal glulamate detection by High Performance Liquid Chromatography (HPLC) at 1 day, 4 days, 7 days and 14 days after the injection. Three models rabbits without intravilreous injection and 3 normal rabbits without any treatment were served as controls. RESULTS: The mean IOP was (19. 32 ± 1.45) mmHg in normal rabbit eyes and (34. 12 ± 2.31) mmHg in experimenlal group. The mean relinal glulamate content of normal rabbit was (1.71 x 10 ± 0.23 x 10 (5)) mmol/g.bul in chronic ocular hypertension eyes, retinal glutamate content was (4. 65 ± 0.12 x 10 (-5)) mmol/g. The latter increased significantly in comparison with the former (1 = 6.318, P = 0. 021). Retinal glutamate content was significant decreased in lire hxpericin-treated eves in comparison with saline-treated eves at various time points (1 = 7.156, P = 0.019 , 1 = 7.933, P = 0.016, 1 = 11.993, P = 0.007, 1 = 5.276, P = 0.034). CONCLUSION: Chrome ocular hvperiension can induce the increase of retinal glutamate content and further result in apoplosis of retinal ganglion cells, lntravitreal injection of hv pericin can decrease the retinal glutamale content.
